Will killing the queen ant get rid of ants entirely?

No...an ant colony that has lost a queen will simply breed a new one and possibly split off into more colonies with more queens. You have to kill the colony or colonies and the best method is to use poison baits that the workers/foragers take back to the nest(s) which destroys the colonies. The best brand that I have found is called Terro ...sold at Walgreens or other home centers, hardware stores, etc... it's mainly borax mixed with a sugar solution that attracts the foragers...they consume it and take it back to the nest to feed the workers which kills the colony. It also depends if the ants are attracted to sugary substances or other foods like starches, breads, etc.. Place some bread out where ants are found and a spill of sugary soda to see which one they are attracted to...then use appropriate baits. Some people soak bits of bread in water/borax mix or mix borax with honey as home remedies but none worked better than Terro IMO.

Best thing is to buy a box of Borax at Your local Grocery Store . Take 1 cup Borax and 2 cup's Sugar and mix and sprinkle anywhere You suspect Ants travel. Place some in a jar lid or other container and leave it near their nests. Easily gets rid of 3 trillion ant's.

We just moved into a new house and it has serious ant problems. I just found the queen crawling across the kitchen and killed her. Will this get rid of the seemingly 3 trillion ants living inside of our home? If not how can I solve the issue?
